Experiments with shortened spadin analogues examined blockade of the TREK-1 potassium channel. PE-22-28 showed activity in channel assays and antidepressant-like behavioral tests in mice. Such tests are tools for exploring mechanisms; they do not establish that PE-22-28 treats depression, improves human cognition or has an acceptable long-term safety profile.
